Some Properties of ¥á-Galactosidase from Bifidobacterium angulatum and Bifidobacterium longum

Abstract
Bifidobacterium angulatum KCTC 3236 (ATCC 27535) and Bif. longum KCTC 3128 (ATCC 15707) were used to study the intracellular ¥á-galactosidase. The enzyme was produced maximally in MRS broth medium containing 1% galactose as a carbon source, with an initial pH 7.0 at 37¡É. The enzyme productivity increased until 17 hours in culture for Bif. angulatum and 20 hours for Bif. longum, and decreased thereafter. Enzymatic hydrolysis of rafinose and stachyose was analyzed by thin-layer chromatography (TLC). Crude ¥á-galactosidase hydrolyzed raffinose and stachvose completely. New oligosaccharides formed by transgalactosylation reaction of the enzyme were observed by TLC.
